PORPHOBILINOGEN, QUANTITATIVE, 24- HOUR URINE

High-Performance Liquid Chromatography (HPLC) with Fluorometric Detection

Includes quantitation of coproporphyrins, uroporphyrins, and intermediate porphyrins (heptacarboxyl, hexacarboxyl, and pentacarboxyl).

Includes liquid chromatography-tandem mass spectrometry (LC-MS/MS) determination of porphobilinogen.

Test Performed At:

Mayo Medical Laboratories   #8562

Specimen Required:

Submit an entire 24-hour urine collection in a urine container that has 5 grams of sodium carbonate added as preservative, supplied by St. Louis Children’s Hospital. Refrigerate specimen during collection and keep specimen protected from light.  Please note collection start and stop date/times on container. Call 314-454-4161 to obtain container.

NOTE:

Please forward a list of medications the patient is currently taking with the specimen.

CAUTION:

PBG and porphyrins are susceptible to degradation at high temperature, at pH <7.0, and on exposure to light. Please be sure to collect specimen in a light-resistant container with preservative that has been added prior to beginning of collection.

Laboratory Notes:

Send 20.0 mL-50.0 mL (minimum 15.0 mL) of urine from a 24-hour urine collection in an amber 60 mL screw-capped bottle, protecting specimen from light. Send specimen refrigerated.

NOTE:

1.

The acceptable preservative must be added to collection container at the start of collection to achieve a pH of >7.0.

2.

DO NOT substitute sodium bicarbonate for sodium carbonate

CAUTION:

PBG and porphyrins are susceptible to degradation at high temperature, at pH <7.0, and on exposure to light. Please be sure to send specimen with preservative that has been added prior to beginning of collection, in a light-resistant container.
